TnaA(tryptophanase from E.coli substrain MG1655)

Tryptophanase could catalyze the reaction from a L-tryptophan to a indole and a pyruvate.


Biological function

TnaA is a tryptophan clastic enzyme from E.coli substrain MG1655, it can decomposes tryptophan into many secondary metabolites like indole and pyruvate, and indole is the precursor of indigo.

Instruction

Tryptophanase is an endogenous enzyme of E. coli, and in most cases, it negatively affects the production of indigo derivatives (Tyrian Purple et al). Thus, in TnaA express process, TnaA deficient strain(TnaA(-)) is usually used as an expression chassis. We constructed the expression sequences in the TnaA(-) E. coli BL21 after codon optimization, and successfully transformed it into TnaA(-) E. coli BL21 to verify the protein expression.
